Model 2450 MHz (PW) exposure to C3H 10T1/2 cells and analysis of transformation frequency (Benz(a)Pyrene and x-rays co-transformaion)

C3H/10T1/2 cells were exposed to 2450 MHz microwaves (PW at 120 pulses/sec with a 83 usec pulse width) for 24 hours at 0.1, 1.0, and 4.4 W/kg under far field conditions using a standard-gain horn antenna. Exposure of RF alone, or RF after benzo[a]pyrene or X-ray initiation did not cause cell transformation. TPA treatment of cells previously irradiated with microwaves, however, resulted in a statistically significant increase in transformation when compared with controls. Microwave treatment before X-ray initiation + TPA promotion treatment resulted in a small increase in transformation frequency over the number of transformants from X-rays + TPA alone. Microwave exposure also reduced the plating efficiency of C3H/10T1/2 cells by 50%, while TPA increased it by 40% in both control and MW-exposed cultures. The authors explain this result by suggesting that MW-induced membrane damage reduces cell adhesion (giving decreased plating efficiency) and is counteracted by the adhesion-promoting effects of TPA.
